Actually using these sealants you are making in effect a flexible self adhesive gasket. If you though tighten the mating surfaces right away when you first apply it you don't leave enough material thickness left to form that gasket and then it'll fail. You instead need to apply the adhesive and only lightly tighten any fasteners and once the material has set, usually 24 hours, go back and tighten the mechanical fasteners.
