Strip: Locking down forms after alignment

This may be a basic question but my instructions are a bit vague.

I'm building on an external strongback. You can see the setup below, with base blocks screwed firmly down, plywood on each block screwed at a single point for adjustment, and forms clamped on the plywood. I've clamped long strips near the sheer line and keel to check alignment.

Do I apply initial strips along the sheer prior to locking everything in place, or lock it down first? I see how a driver could yank stuff out of place, so I figured I'd ask rather than guessing.
